测定意义：

β-GAL(EC 3.2.1.23)广泛存在于动物、植物、微生物和培养细胞中，能够催化β半乳糖苷化合物中β半乳糖苷键水解，此外还具有转半乳糖苷的作用。β-GAL不仅可为植物的快速生长释放储存的能量，还能在正常的多糖代谢、细胞壁组分代谢以及衰老时细胞壁降解过程中催化多糖、糖蛋白以及半乳糖脂末端半乳糖残基的水解，释放自由的半乳糖。
测定原理：

β-GAL 分解对-硝基苯-β-D-吡喃半乳糖苷生成对-硝基苯酚，后者在400nm 有*吸收峰，通过测定吸光值升高速率来计算β-GAL 活性。
自备用品：

可见分光光度计、台式离心机、水浴锅、可调式移液器、1mL 玻璃比色皿、研钵、冰和蒸馏水。
