苏木素是一种碱性染料，同媒染剂（常用的是三价的铁或铝的盐）一起使用，能够使细胞核染色。苏木素和氧化苏木素都没有染色能力，但与媒染剂结合形成色淀（lake），这种色淀具有染色能力。核、染色体、着丝粒、中心体、线粒体、髓鞘等可被染成蓝色以至黑色。与伊红合用的苏木素-伊红双重染色法是最基本的一般组织的染色法，苏木素染色原理如下：
苏木素和伊红在组织学上被经常使用，碘液不同，这是一种*染色街。其他常用的含有苏木素的染色剂有磷钨酸苏木素染色剂，也就是磷钨酸与苏木素的混合物。苏木素染色剂经常为组织的研究使用。明矾和三价铁盐常常被用作媒染剂，展示核和细胞质结构。这些媒染剂的原理都是形成媒染剂－染料－组织复合体，从而显示颜色。
苏木素染色内质呈淡蓝黑色，外质收缩，剩下部分不着色或色泽更浅。圆形细胞核呈蓝黑色。居中的核仁呈黑色小圆点。核膜较薄，其内侧缘均匀，整齐地排列着一层细小的染色质粒。核仁与核膜之间可见到核网。被吞噬的红细胞呈蓝黑色采用苏木素染色液进行免疫染色后的复染，操作步骤简便，操作时间短。可以操作步骤简便，操作时间短或培养细胞的染色，或与免疫组织化学、免疫细胞化学、免疫荧光、原位杂交等配合使用。使用苏木素染色液染色后还可以进行免疫染色或其它染料的复染。苏
苏木素染色原理很简单，苏木素染色法也是常用的一种染色方法。木素染色后的细胞核呈现蓝色，细胞浆呈现粉红色或红色。染色液也可以重复使用，直至认为效果不佳时再换用新的染色液。
